چکیده

Elevated levels of DDT and other organochlorine pesticides have been associated with spontaneous abortion and preterm birth in several species, including humans (Delong et al. 1973; Hart et alo 1971; O'Leary et alo 1970; Saxena et al. 1980, 1981; Wassermann et al. 1982). Despite the prevalence of organochlorine pesticides in our environment, a mechanistic basis for this association has not been explored. Furthermore, while DDT has been associated with inhibition of calcium ATPases (Ghiasuddin and Matsumura 1981; Treinen and Kulkarni 1986), altered gap junctional communication (Madhukar et al. 1983; Davidson et al. 1985), and electrophysiological changes (Schefczik and Buff 1984; Beeman 1982), all of which could affect the excitation-contraction process characteristic of smooth muscle, direct effects of DDT on uterine smooth muscle have not been reported. This study was initiated to assess the direct effects of o,p'-DDT (an estrogenic isomer present in the technical grade preparation) on pregnant rat uterine tissue.

Investigation prostaglandin of the role of estrogenic action and E2 in DDT-stimulated rat uterine contractions ex vivo

Previous work in our laboratory showed that o,p'-DDT increases the frequency of rat uterine contractions in vitro. The present study investigated whether this response was related to prostaglandin E 2 (PGE2) release from the uterine strips or to the estrogenicity of o,p'-DDT. Contraction frequency was evaluated by recording isometric spontaneous contractions in longitudinal uterine strips from ...

Characterization of o,p'-DDT-stimulated contraction frequency in rat uterus in vitro.

Exposure to organochlorine pesticides, including DDT, has previously been associated with premature birth. Using an improved protocol to characterize dose and time dependent responses, the present report extends a preliminary finding by this laboratory that o,p'-DDT directly stimulates uterine contractility. Contraction frequency was determined in longitudinal uterine strips from pregnant rats ...

Chronic levothyroxine and acute T3 treatments enhance the amplitude and time course of uterine contractions in human.

This study compares the functional consequences of levothyroxine (T4) treatment during pregnancy as well as the acute affects of triiodothyronine (T3) on spontaneous uterine contractile activities observed in vitro. Uterine biopsies were obtained from consenting women undergoing elective caesarean at term (n = 28). Spontaneous contractile activities from T4-treated pregnant women (n = 8) were c...

Levothyroxine treatment generates an abnormal uterine contractility patterns in an in vitro animal model

Objective Abnormal uterine contraction patterns were recently demonstrated in uterine strips from pregnant women treated with Levothyroxine (T4). These abnormalities were correlated with an increased risk of C-section delivery and associated surgical complications. To date, no study has investigated whether uterine contractility is modified by hypothyroidism or T4 treatment. Herein, we analyze ...

Stimulation of contraction of pregnant rat uterus in vitro by non-dechlorinated and microbially dechlorinated mixtures of polychlorinated biphenyls.

A previous study of six polychlorinated biphenyl (PCB) congeners showed that PCBs with four or fewer chlorines and ortho substitution stimulate uterine contraction frequency in vitro, whereas congeners with a greater number of chlorines or non-ortho substitution are inactive in vitro. We tested the hypothesis that PCB mixtures stimulate uterine contractions in a manner inversely related to the ...
